Calculated Risk: AIA: Architecture Billings Index increases in ...

The American Institute of Architects (AIA) reported the September ABI score was 51.6, up from the mark of 50.2 in August. This score reflects an increase in demand for design services (any score above 50 indicates an increase in billings). The new projects inquiry index was 57.3, compared to a mark of 57.2 the previous month.

?Going back to the third quarter of 2011, the multi-family residential sector has been the best performing segment of the construction field,? said AIA Chief Economist, Kermit Baker, PhD, Hon. AIA. ?With high foreclosure levels in recent years, more stringent mortgage approvals and fewer people in the market to buy homes there has been a surge in demand for rental housing. The upturn in residential activity will hopefully spur more nonresidential construction.?

This graph shows the Architecture Billings Index since 1996. The index was at 51.6 in September, up from 50.2 in August. Anything above 50 indicates expansion in demand for architects' services.

This increase is mostly being driven by demand for design of multi-family residential buildings - and this suggests there are more apartments coming (there are already quite a few apartments under construction). Note: This includes commercial and industrial facilities like hotels and office buildings, multi-family residential, as well as schools, hospitals and other institutions.

According to the AIA, there is an "approximate nine to twelve month lag time between architecture billings and construction spending" on non-residential construction. This suggests increase in CRE investment next year (it will be some time before investment in offices and malls increases).

Internet Marketers Must Listen Before They Speak - Search Engine ...

As marketers, we spend a lot of time thinking about our message, as well we should. Our job is to tell prospective customers how we can help them, so we focus on what we say. The Internet has turned the tables on marketers, however. Time was that marketing messages were conceived by extremely creative people to get attention for products, but now, listening to what customers say and do are just as important as creativity. Old-time marketers have always listened to a few customers in focus groups, so you can think of the Internet is the biggest focus group of all time.

All Internet marketing is more successful when you listen before you speak. And if you listen before you speak again. You must listen to what your customers say and you must watch what they do. Only by seeing what works and what doesn't do you have a chance of persuading them to believe your marketing message.

Take social media as an example. A proven social media practitioner would advise you that before you engage in social media that you should first listen to the conversation that is already out there about your products and your industry. Failing to do so is like walking into a cocktail party where you don't know anyone, not listening to what is going on, and just starting to talk to no one in particular.

Working with a firm that employs listening technology as part of itsmodus operandi will get your social media marketing off on the right foot. Merely going at this like an ad agency with a clever person who comes up with ideas in a vacuum just won't achieve the same success, in my experience.

But it's not just social media. With search, you must wait for the searcher to initiate-so you are listening first. With banner ads, you can test them to see which ones work better than others. With e-mail, you can test response with small mailing lists before sending the winning version to the main list.

With all forms of Internet marketing, listening to what customers say and watching what they do are the key methods for fine-tuning your message until it resonates with your audience.

This might sound like a lot of work. It is.

Instead of just thinking up your message and delivering it, you are constantly tweaking and changing what you say based on the customer response. And it is time-consuming, honestly. It undoubtedly is less work to come up with just one message and deliver it, than it is to come up with dozens of messages, constantly tinkering to get your final one.

But you're not trying to minimize the amount of work to come up with a message. You're trying to maximize response. And there's no way that your first message will be as good as your 50th. So, even though it seems like coming up with 50 messages is more work than coming up with one (because it is), it ends up being easier to get results this way. So, if you can guarantee getting good results one way, but not the other, which one is less work in the long run?

Ask yourself: If you aren't listening to your customers to fine-tune your messaging, just what are you listening to?

Here's how to take an HTPC that's driving you crazy and whip it back into shape so you can enjoy the show.

I love my home theater PC (HTPC), but it's not exactly perfect. For a long time, XBMC wouldn't download art, plot data, or cast information. The interface is difficult for my friends, who are used to cable TV. Using a keyboard and mouse on the couch sucks sometimes. We're willing to bet you've noticed this with your HTPCs and just deal with it, or you've held off building one because of similar annoyances. You don't have to settle for a second-rate experience. In this post we'll show you how to make your HTPC work the way you want it to.

My HTPC Won't Play Some of My Files

Nothing's worse than sitting down to watch a new episode of your favorite show only to find out that the file is in a format your chosen media center can't play. You have two options on how to fix this:

  • Get a media center application that transcodes video on the fly. A few apps, most notably Plex, Subsonic, and PS3 Media Server all do this seamlessly. Transcoding makes sure you can watch any type of media on any device you happen to be using, including smartphones and tablets. You can either transcode on the source machine (if the files are stored on a home server or other computer) or your HTPC itself. Pick whichever computer has a faster processor to do the decoding.
  • The Six Biggest Media Center Annoyances (and How to Fix Them)Convert all of your media to a single file format that's compatible with everything. This method requires a bit more work up front, but has a potential for less problems when you're actually trying to watch your videos. Basically, you just need to go through your entire library and convert all of your media to a common format. You can then set up a system that converts it as soon as you download or rip it, so you don't have to do the extra legwork yourself. If you use uTorrent, we've shown you how to roll video conversion into your download process easily. Alternatively, you can use previously mentioned DropFolders with Handbrake, which will watch a download folder and convert whatever comes through it automatically, without you lifting a finger.

Whichever route you choose, the end result should be the same?all of your media should play seamlessly with whatever tool you prefer to watch it. The transcoding method tends to lock you into a specific app, but if you're okay with that, or you know that app is available for all of your devices and set-top boxes, then it's a bit more futureproof. Converting your music and movies on your own takes a lot more work up front, but leaves one fewer link in the chain that could cause problems when you're trying to watch your movies.

This is the issue that actually inspired this post. For months, my movies and TV shows were playable in XBMC, but it wouldn't download posters, DVD cover art, or background information for any of my files. The fix turned out to be somewhat simple: my filenames and directory structure made sense to me, but they made no sense at all to XBMC. We've talked about how to organize your media for XBMC before, and while normally the process is automatic, it didn't work for me. In my case, adding a whole drive with a bunch of unorganized folders and mixed media just caused XBMC to give up. All it required was a bit of organization.

The main change I had to make was to use a supported naming convention for my TV shows and movies so XBMC (or Plex, or anything else) could recognize what they were. Organizing movies and TV shows into separate folders helped a lot too. If you have a lot of files to rename (like I did,) grab an app like The Renamer (Windows) or NameChanger (OS X) to help speed up the process.

Home Theater PCs Are Too Expensive

Building an HTPC doesn't have to be an expensive proposition. We've shown you how to build one for less than $500, and another for around $200, but you can go even lower than that if you wish.

Your cheapest option is probably to grab a $99 Apple TV (less if you get a refurb!). It's a great set-top box to begin with, but you can take it up a notch by jailbreaking it and installing XBMC or Plex (although the current gen isn't jailbreakable, the 1G and 2G models are.) XBMC will let you play any video type, stream from other computers, and more that the Apple TV won't do on its own. Need more choices? Where Do I Store My Movies?

You've probably heard us talk about streaming from a home server, from another computer, and even from smartphones, but where should you actually store your media? The best place to store your movies depends on how you plan to watch them. Here are a few scenarios:

  • Store and watch movies on your HTPC. This is obviously the easiest method: if you have a big enough hard drive in your HTPC, you can just put the movies there. However, if you need more than one hard drive, need to transcode videos, or want your HTPC to download stuff in the background, you'll need a pretty powerful HTPC, or you'll lose some performance while all that is going on. It's probably cheapest, but it's also a single point of failure, and it too needs to be on all the time. Also, if you have something like a jailbroken Apple TV, this isn't an option because it doesn't have internal storage.
  • Store and transcode movies on your main PC, watch them on your HTPC. If you need a place to store, transcode, and download media, you can always do this from your main computer. Your powerful computer can handle things like converting files and automatically downloading movies and TV shows as soon as they air, then you can stream them to your HTPC for watching. It's a power-hungry setup, and your desktop PC will have to be turned on pretty much all the time, but it's an option that doesn't require another computer.
  • Store movies on a NAS, watch them on your HTPC. This is our suggestion, mostly because your NAS, whether you buy one or roll your own, is designed for low power consumption. This is good, since you'll need to keep it on all day, which you probably don't want to do with your desktop computer. A NAS gives you multiple hard drives for massive storage, RAID for redundancy, and enough power to do all that renaming, transcoding, and even downloading for you?without such a harsh strain on your energy bill. We've tackled this topic before, so it's not terribly difficult to get your HTPC in order so anyone can flop down on your couch and use it. Your first step is to get and set up a remote for your HTPC to put people at ease?after all, you may be comfortable with just a keyboard and mouse, but many people aren't, so keep it stashed away unless you need it. Even better, grab your tablet or an old smartphone and use these iOS or Android apps to control your HTPC from anywhere in the house.

That takes care of the remote problem, but your HTPC's interface could probably use some work too, right? You can tweak a few settings in XBMC to make it more friendly to non-geeks right out of the box, though a new skin can also be helpful. That doesn't mean you're out of options. There's always Hulu and Netflix, or you could drop a TV Tuner into your DVR and attach an HD over-the-air antenna to it to watch and record live television. Romney plays it safe. Smart move?

There were times during this last debate when I almost thought I could hear the words of Mitt Romney?s advisers playing in his head:

?Look, big guy, you?re on track to win this thing. What they want to see tonight is a calm, confident leader, unthreatening, informed, unruffled. So don?t get up in Obama?s grill. Bring the conversation back to the economy when you can, and be the reasonable, credible Commander-in-Chief the voters want.?

That was clearly the strategy Romney executed Monday night. He was at pains to agree with President Barack Obama on matters ranging from the deployment of drones to standing with Israel in the face of an attack. He drew attention to Obama?s sharp criticisms by saying ?attacking me is not an agenda??a line he repeated later. He stepped back from a frontal assault on the administration?s confusion over Libya, speaking more broadly about the Middle East, observing, ?We can?t kill our way out of this mess,? and later talking about the need to promote ?gender equality.? (I wondered at this point whether the campaign had focused Romney?s attention on the baseball and football games competing for the audience, raising the likelihood that a greater percentage of the audience might be female).

Indeed, there were moments when Romney seemed to be channeling the presence of another ex-governor whose need as a presidential candidate was to reassure.

But unlike Ronald Reagan, Mitt Romney did not need to prove that he is something other than a threat to the peace.

Rather, his challenge was to stand?or sit?face to face with the incumbent president and demonstrate that he could credibly argue matters of state, in the face of a debate foe determined to thrust and spar at every opportunity. Without question, Obama came into this last debate knowing that his presidency is hanging by a thread, in large measure due to his remarkably weak performance in the first debate. There was no opportunity he let pass. At one point, he pivoted from the killing of Bin Laden to a conversation he had with a girl whose father had died on Sept. 11, 2001. (I could not help but wonder what the people who charged George W. Bush with politicizing Sept. 11 might have to say about that).

What happened in this debate, then, was what often happens?in sports and in politics?when someone plays not to lose. Obama clearly dominated the debate, as the instant polls suggested. But now a different question arises. What if the premise of the Romney strategy was wrong?

Indeed, what if the premise of every pre-debate assessment?that this last debate was on a matter peripheral to voters? central concerns?is wrong? What if a significant sector of the electorate is still weighing the respective claims of the contenders to be in command when a threat to American security arises?

If that?s the case (and we will not know for several days), it may turn out that playing it safe may have been the least safe course of all.

U.N. sees "lost opportunity" in global crime fight

VIENNA (Reuters) - More than 100 countries lost an opportunity to improve the fight against cross-border organized crime by failing to agree a new monitoring measure at a major international meeting last week, a United Nations agency said on Monday.

Disagreement over how the effort would be funded prevented a deal on introducing a "peer review mechanism" designed to increase the effectiveness of the U.N. Convention on Transnational Organized Crime (UNTOC), which has so far been ratified by 172 nations.

"This is a period of austerity. As a result, states appear wary of taking on new obligations," David Dadge, spokesman for the United Nations Office on Drugs and Crime (UNODC), said in a statement about the outcome of the October 15-19 meeting in Vienna.

The convention forms the legal basis for international cooperation against global crime, which UNODC estimates at $870 billion annually.

The inability to agree "can be characterized as a lost opportunity to further enhance the effective implementation of the Convention, as well as improve the way we fight transnational organized crime", Dadge said, giving no details.

If it had been adopted, countries would have been reviewed to see how they were meeting their obligations under the convention and to identify gaps in national legislation.

UNODC Director General Yury Fedotov last week stressed the importance of the measure, telling delegates: "We cannot succeed against transnational crime unless every nation is bound in cooperation through the convention and the review process."

Dadge said the meeting had still made "considerable progress" in many areas, for example regarding trafficking of firearms and smuggling of migrants.

Bell dealt to D-backs, who trade Young to Oakland

PHOENIX (AP) ? Heath Bell became the latest player jettisoned by the Miami Marlins when he was dealt Saturday to the Arizona Diamondbacks, who also acquired infielder Cliff Pennington from the Oakland for outfielder Chris Young.

Arizona obtained Pennington and minor league infielder Yordy Cabrera from Oakland for Young and cash, then sent Cabrera to the Miami Marlins for Bell. The Marlins will pay $8 million of the remaining $21 million Bell is owed.

Earlier in the day, Arizona exercised a $6.5 million option on closer J.J. Putz.

The 35-year-old Bell has 151 saves the past four seasons. He signed a $27 million, three-year contract with Miami last offseason but lost his closer's job in July. He finished with 19 saves in 27 chances and a 5.09 ERA in in 73 games.

After failing to contend in the first season of their new ballpark, the Marlins traded former NL batting champion Hanley Ramirez to the Los Angeles Dodgers in July and sent pitcher Anibal Sanchez and infielder Omar Infante to the Detroit Tigers.

Arizona general manager Kevin Towers said Bell provides a right-handed power arm to help set up Putz and Pennington adds the team experience at shortstop and second base.

Towers, who was general manager in San Diego when Bell was the setup man for Trevor Hoffman, believes the pitcher will benefit from a return to the NL West.

"I think he's excited to kind of be able to clean the slate," Towers said.

Bell contract calls for $9 million in each of the next two seasons and includes a $9 million club option for 2015. Miami will pay $1.5 million next year, $3.5 million in 2014 and the $3 million deferred signing bonus he is owed.

Young, Arizona's center fielder for six seasons, was the odd man out in a crowded outfield with the team expecting young Adam Eaton, called up from Triple-A Reno late this season, to play center and be the Diamondbacks' leadoff hitter. But Towers said the job is automatically Eaton's. Gerardo Parra and A.J. Pollock also will compete for the spot.

Young will be reunited with Bob Melvin, who was his manager in Arizona.

"It caught me off guard a little bit, I'm not going to lie," Young said. "It's all settled in a couple hours now. I got an opportunity to talked to Billy (Beane) and got to talk to BoMel. I'm excited to help. It's a new opportunity, a fresh start for me personally."

Pennington, 28, played in 125 games for the A's. He had 93 appearances at shortstop but was shifted to second base in last August. A switch hitter, Pennington hit .215 last season, including .168 against left-handers, and had 28 RBIs.

"He had a down year. That happens. Guys have down years," Towers said. "Heath had a down year. That's why we were able to make these deals."

He said he believes Pennington can be an everyday shortstop plus fill in behind Aaron Hill at second. The team also has Willie Bloomquist and John McDonald at short.

Towers said that Bell may have been uncomfortable with his high-profile signing in the Miami makeover.

"Maybe a lot of it had to do with the pressure, his first big contract," Towers said, adding that Bell "was really the first big signing" the Marlins had in the offseason.

The Arizona general manager believes his bullpen ? led by Putz, David Hernandez, Brad Ziegler and now Bell ? "is as good a bullpen as there is if not in the National League but in baseball."

With considerable money invested in the bullpen, Towers said it's particularly important to have a strong group of relievers while Arizona's young starting rotation. He said Arizona still needs a late-inning left-handed reliever and perhaps a veteran starter.

This could be a sign Beane, the A's general manage,might try to move center fielder Coco Crisp, who signed a $14 million, two-year contract in January that includes a $7.5 million club option for 2014 with a $1 million buyout. Beane, however, said he plans to keep Crisp, though he realized there would be speculation otherwise.

" I really like our outfield," Beane said. "Everybody knows how important this guy is to this team and also has the benefit of being a personal favorite of mine."

Oakland already acquired shortstop Stephen Drew from the Diamondbacks this season and he helped them down the stretch. The A's overtook Texas on the final day of the regular season to win the AL West and returned to the playoffs for the first time since 2006. They lost in five games to the World Series-bound Detroit Tigers.

Beane said the day after the season ended he planned to keep his young team intact as much as possible. Last offseason, Oakland traded three top pitchers: Trevor Cahill to Arizona, Gio Gonzalez to Washington and closer Andrew Bailey to Boston.

The 29-year-old Young, a popular player in the Arizona clubhouse, has had three 20-homer, 20-steal seasons. He injured his right shoulder crashing into a wall while making a catch early this season and struggled at the plate after. A quadriceps injury in early September sent Young to the bench in favor of Eaton.

Forest fires linked to high temperatures two years before

ScienceDaily (Oct. 19, 2012) ? A study led by some University of Barcelona researchers analyses the impact of interannual and seasonal climate variability on the fires occurred in Catalonia last summer. The study concludes that summer fires, related to summer climate conditions, are correlated with antecedent climate conditions, especially winter and spring ones with a lag time of two years. The results suggest that precipitation and temperature conditions regulate fuel flammability and fuel structure. According to the correlations observed, the study provides a model to produce long-term predictions.

The study, published in the journal Climatic Change, comes out of the doctoral thesis of the researcher Marco Turco, directed by the UB researcher Maria del Carme Llasat, co-author of the article. From 1983 to 2007, period analysed in the study, more than 16000 fires events were recorded and the total burned area was more than 240000 hectares, around 7.5 % of Catalonia. The work develops a statistical analysis of these fires and shows that, from a climate point of view, according to Maria del Carme Llasat, "is possible to develop a model that gives us an estimation of the number of fires and the extension of the burned area related to monthly average temperature and rainfall. We developed a simple regression model which includes the influence of spring-summer climate conditions of the studied year, but specially other variables which are determinant, although they do not seem to."

The established correlations allow us to prove that, for example, low minimum temperatures in winter and summer contribute to an increase in the number of fires. However, the extension of the burned area highly depends on the winter months' rainfall, and in both cases on the winter-spring temperatures with a time lag of two years.

Specifically, the number of fires is correlated with the minimum temperature in the period February-June of the previous two years, and the number of burned hectares is correlated with the maximum temperature in the period March-April of the previous two years. "Even if it is not confirmed yet, this relation with climate data of two previous years has to do with the vegetation cycle of the region studied," explains Llasat, head of the group Metereological Hazard Analysis (GAMA). This two years relation has also been observed on other works developed in the Mediterranean area.

Fires are a complex processes associated with factors of different origin, such as climate and weather, human activities and vegetation conditions. As a first approximation a concept model was proposed suggesting that climatic processes act as top-down controls on the regional pattern of fire controlling both fuel moisture (fuel flammability) and nature and availability of fuel (fuel structure). Catalonia can be classified as an ecosystem in which climate acts in two main ways: antecedent climate regulates the fuel amount and its continuity, whilst current climate (for example, drought) promotes the fuel load.

Adopting the view that climate is the main controlling factor of the interannual variability of fire, on this work the links between climate and fire variability are analysed using the high resolution (20 km x 20 km) gridded data set, called Spain02. This data set is produced by the Spanish Metereological Agency (AEMET). Accurate data for fire occurrence and burned are were obtained from the Forest Fire Prevention Service of the Generalitat de Catalunya (SPIF). The data consists in the characteristics of 16753 fire events occurred in Catalonia during the 25 years period studied. Although fires in this region occur throughout the year, about 60% of the fires occur in summer, from June to September, which amounts to be about 86% of the annual burned area.

Model applications

In the study a simple regression model is presented that links the variability of summer fires in Catalonia to climatic variables, and produces reliable out-of-sample seasonal and climatic predictions.

Llasat points out: "With this model we can make reliable predictions about the impact of climate variability on summer fires, but it is not the right model to be used for risk evaluations of prompt fires. In order to do that we have other resources, such as the one used by the SPIF and developed in the UB, which take into account other variables, for example the wind."

"We performed a first out-of-sample prediction that in general fits well, except for the 1994 big fires. We also studied this year meteorologically and we know that it was exceptional," explains the researcher. The model explains up to 76 % of the variance of the burned area and up to 91 % of the variance of the number of fires.

UB researchers are working in order to use the model to estimate fire response to different climate change scenarios, assuming that climate, vegetation, humans, fire interactions will not change significantly.

SOUTH BEND, Ind. (AP) ? Theo Riddick is Notre Dame's smallest running back but Brigham Young learned he's got no shortage of power.

The 5-11, 200-pound Riddick ran for a career-high 143 yards against a Cougars defense that entered the game third in the nation against the rush. More importantly, Riddick sparked a ground game that ground out 270 yards, with his housemate, Cierre Wood, getting 114 yards and George Atkinson III scoring the go-ahead touchdown as the Irish rallied for a 17-14 win Saturday.

"As a team, this was special," Riddick said. "We showed that we can run the ball."

He got no argument from Notre Dame coach Brian Kelly.

Riddick "just ran really hard, north and south, physical," Kelly said. "That gives energy for everybody. The o-line sees a guy who's really pounding it in there. He got us that energy we needed."

Notre Dame (7-0), off to its best start in a decade, has a big game ahead against No. 10 Oklahoma next week, although it looked for a while the importance of that game was in jeopardy. The Cougars (4-4) fell to 0-3 on the road as they surrendered a season-high 270 yards rushing.

The Cougars drove to the Notre Dame 31 late, but Riley Nelson was sacked for a 9 yard loss. Moments earlier Cody Hoffman was wide open for what would have probably been a touchdown, but Nelson's pass wasn't close.

"We were one or two plays short from being able to come away with a victory," BYU coach Bronco Mendenhall said.

Riddick had runs of 55 and 27, the two longest rushes of his career. Neither was easy. On the 55-yard run from the 37-yard line, he ran into a pile at the line of scrimmage, put his hand down to keep his balance and then broke out of the pack before being caught by cornerback Jordan Johnson at the 8-yard line.

"Just run and run until you can't no more," Riddick said. "And I got a little gassed at the end."

The Irish then ran three straight running plays for a gain of one yard and had to settle for a 24-yard field goal by Kyle Brindza to cut BYU's lead to 14-10 late in the third quarter. On his 27-yard run late in the second quarter, Riddick broke two tackles and carried three BYU defenders for the final 10 yards to the BYU 14. But the drive stalled and Brindza missed a 28-yard field goal wide right.

Riddick said it was exciting to see the Irish backs come up so big.

"We have some talent back there, so why not give us the ball?" he said.

BYU linebacker Brandon Ogletree said the Cougars didn't tackle well enough.

"There were way too many times where we had a dude down in the backfield, in a hole, and we let him spring one for 12, 20, 40 yards, so that's not acceptable," he said.

After going 17 quarters without giving up an offensive touchdown, Notre Dame gave up two in a span of 2:18 as Nelson threw a 6-yard scoring pass to a wide-open Cody Hoffman in the end zone and a 2-yard pass to Kaneakua Friel after an interception by Kyle Van Noy.

Key player in Parkinson's disease neuron loss pinpointed

ScienceDaily (Oct. 19, 2012) ? By reprogramming skin cells from Parkinson's disease patients with a known genetic mutation, researchers at the Salk Institute for Biological Studies have identified damage to neural stem cells as a powerful player in the disease. The findings, reported online October 17th in Nature, may lead to new ways to diagnose and treat the disease.

The scientists found that a common mutation to a gene that produce the enzyme LRRK2, which is responsible for both familial and sporadic cases of Parkinson's disease, deforms the membrane surrounding the nucleus of a neural stem cell. Damaging the nuclear architecture leads to destruction of these powerful cells, as well as their decreased ability to spawn functional neurons, such as the ones that respond to dopamine.

The researchers checked their laboratory findings with brain samples from Parkinson's disease patients and found the same nuclear envelope impairment.

"This discovery helps explain how Parkinson's disease, which has been traditionally associated with loss of neurons that produce dopamine and subsequent motor impairment, could lead to locomotor dysfunction and other common non-motor manifestations, such as depression and anxiety," says Juan Carlos Izpisua Belmonte, a professor in Salk's Gene Expression Laboratory, who led the research team. "Similarly, current clinical trials explore the possibility of neural stem cell transplantation to compensate for dopamine deficits. Our work provides the platform for similar trials by using patient-specific corrected cells. It identifies degeneration of the nucleus as a previously unknown player in Parkinson's."

Although the researchers say that they don't yet know whether these nuclear aberrations cause Parkinson's disease or are a consequence of it, they say the discovery could offer clues about potential new therapeutic approaches.

For example, they were able to use targeted gene-editing technologies to correct the mutation in patient's nuclear stem cells. This genetic correction repaired the disorganization of the nuclear envelope, and improved overall survival and functioning of the neural stem cells.

They were also able to chemically inhibit damage to the nucleus, producing the same results seen with genetic correction. "This opens the door for drug treatment of Parkinson's disease patients who have this genetic mutation," says Belmonte.

The new finding may also help clinicians better diagnose this form of Parkinson's disease, he adds. "Due to the striking appearance in patient samples, nuclear deformation parameters could add to the pool of diagnostic features for Parkinson's disease," he says.

The research team, which included scientists from China, Spain, and the University of California, San Diego, and Scripps Research Institute, made their discoveries using human induced pluripotent stem cells (iPSCs). These cells are similar to natural stem cells, such as embryonic stem cells, except that they are derived from adult cells. While generation of these cells has raised expectations within the biomedical community due to their transplant potential -- the idea that they could morph into tissue that needs to be replaced -- they also provide exceptional research opportunities, says Belmonte.

"We can model disease using these cells in ways that are not possible using traditional research methods, such as established cell lines, primary cultures and animal models," he says.

In this study, the researchers used skin fibroblast cells taken from Parkinson's disease patients who have the LRRK2 mutation, and they reprogrammed them to iPSC stem cells and developed them into neural stem cells.

Then, by using an approach to model what happens when these neural stem cells aged, they found that older Parkinson disease neural stem cells increasingly displayed deformed nuclear envelopes and nuclear architecture. "This means that, over time, the LRRK2 mutation affects the nucleus of neural stem cells, hampering both their survival and their ability to produce neurons," Belmonte says.

"It is the first time to our knowledge that human neural stem cells have been shown to be affected during Parkinson's pathology due to aberrant LRRK2," he says. "Before development of these reprogramming technologies, studies on human neural stem cells were elusive because they needed to be isolated directly from the brain."

Belmonte speculates that the dysfunctional neural stem cell pools that result from the LRRK2 mutation might contribute to other health issues associated with this form of Parkinson's disease, such as depression, anxiety and the inability to detect smells.

Finally, the study shows that these reprogramming technologies are very useful for modeling disease as well as dysfunction caused by aging, Belmonte says.
